Abstract: The assumptions implicit in the deductions made from the pH-dependence of rate measurements of enzyme-catalyzed reactions are summarized, and the limitations of such determinations are discussed nonalgebraically. The following types of pH-profile are considered (in order of increasing utility): pH-"activity" curves at fixed [S] o;pH-dependences of kcat, Km, kcat/Km, and Ki; pH-dependences of kmodification (by specific reagents) and of competitive labeling (by nonspecific reagents); pH-dependences of elementary steps; and the direct observation of a titrating group.
Few studies were made on the intrinsic and co-intrinsic vowel characteristics in neurologic dysarthrias. This work evaluates these parameters in a group of 4 subjects with cerebellar dysarthria suffering from a cerebellar degenerative syndrome and a group of 4 subjects with Parkinson disease suffering from parkinsonian dysarthria. These subjects were compared with 10 control subjects. An intensive speech treatment inspired from the Lee Silverman voice treatment (LSVT) was applied to one of the parkinsonian subjects of the study, with assessment...
We continue our previous work on dynamic "intrinsically random" systems for which we can derive dissipative Markov processes through a one-to-one change of representation. For these systems, the unitary group of evolution can be transformed in this way into two distinct Markov processes leading to equilibrium for either t--> + infinity or t--> - infinity. To lift the degeneracy, we first formulate the second principle as a selection rule that is meaningful in intrinsically random systems. For these systems, this excludes a set of unrealizable s...
The primary purpose for the in vitro system described here was to provide an alternative method to intrinsic isotopic labeling for determining exchangeability of intrinsic food mineral with extrinsic inorganic mineral or one of its isotopes. In this system, foods or food mixtures extrinsically tagged with an isotope of the mineral of interest are incubated successively in media simulating peptic or pancreatic digestion. Radioactivity and mineral measurements are made on a)the mixture before digestion and after b)peptic and c)successive peptic p...
The present experiments were designed to analyze the influence of copper and iron ions on the process of decomposition of S-nitrosocysteine (cysNO), the most labile species among S-nitrosothiols (RSNO). CysNO fate in buffer solution was evaluated by optical and electron paramagnetic resonance (EPR) spectroscopy, and the consequences on its vasorelaxant effect were studied on noradrenaline-precontracted rat aortic rings. The main results are the following: (i) copper or iron ions, especially in the presence of the reducing agent ascorbate, accel...
Capillary wave fluctuations smooth out the structure of fluid interfaces, making difficult the detailed analysis of the interfacial structure. Most computer simulation investigations performed to date have focused on the computation of average density profiles, ignoring the characterization of the intrinsic structure of the interface. Recent theoretical developments have reversed this situation, making possible the detailed investigation of the interfacial intrinsic structure at an unprecedented level of detail. In this article we investigate v...
Granulocytes are important cells of inflammation and cellular thrombolysis. They produce urokinase (u-PA) and chloramines. In this study, u-PA/chloramine-mediated fibrinolysis is imitated in a microtiter-plate. Seventy-five microliters plasma are incubated with 50 microL 50% Pathromtin SL, 6% BSA, and 38 mM CaCl2 for 30 minutes (37 degrees C). Then, 50 microL 10 mM chloramine-T in PBS are added. After 30 minutes (37 degrees C), 50 microL 0, 100, or 10 IU/mL u-PA in 6% BSA-PBS are added and the turbidity is determined at 405 nm after 0, 3, or 16...
The traditional view of the pathophysiology of asthma is that its characteristic features are secondary to a major allergic or immunological dysfunction. However, this does not explain intrinsic asthma, which can occur in the absence of atopy. An alternative view is that an abnormality in the airway smooth muscle cell, which is capable of producing inflammatory, immunological and growth factors as well as molecules, which facilitate interaction with inflammatory cells, is the primary or instigating event. Evidence is rapidly accumulating that t...
Abstract The issue of intrinsic bistability in resonant tunneling devices is examined analytically and quantitatively. The results indicate that the putative argument for intrinsic bistability is actually an argument for intrinsic tristability. If a third stable configuration really exists, it might be hard to access experimentally although its existence could have consequences for high-speed device applications. If an intrinsic bistable regime is ipso facto a tristable regime, then an experimental search for a third stable configuration could...
